PTPRC,即蛋白酪氨酸磷酸酶受体C型,它是一种跨膜蛋白酪氨酸磷酸酶,广泛表达于T淋巴细胞、B淋巴细胞、自然杀伤细胞(NK细胞)和巨噬细胞等免疫细胞中。PTPRC通过去磷酸化作用调节多种信号传导途径,包括但不限于细胞生长、分化、有丝分裂和致癌转化等细胞过程。它参与调控的信号通路主要有T细胞受体(TCR)和B细胞受体(BCR)的信号传导,通过与抗原受体复合物直接相互作用,或激活Src家族激酶,对抗原受体信号转导进行调节。此外,PTPRC还抑制JAK激酶,作为细胞因子受体信号转导的调节者。在生物学意义上,PTPRC对维持免疫系统的正常功能至关重要,其异常表达或功能失调与多种自身免疫疾病和癌症的发生发展有关。

Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
