E7蛋白具有转化活性和反式激活活性。 结构为两个功能区域:一个是N末端区域,含有保守的Cys-X-X-Cys(CXXC)结构,用于与细胞中的特定蛋白质相互作用;另一个是C末端区域,包含保守的LXCXE序列,用于与细胞中的Rb蛋白(Retinoblastoma protein)结合。E7蛋白通过与细胞中的Rb蛋白结合,抑制Rb蛋白的功能,导致细胞周期的失控和异常增殖。这会使细胞失去正常的生长抑制机制,促进细胞转变和癌变。E7靶点及其与Rb蛋白相互作用的研究已成为HPV相关癌症治疗和疫苗研发的重要方向,是HPV感染中的关键蛋白之一,它与细胞周期调控、转录调控和细胞增殖等过程密切相关。
