集落刺激因子

集落刺激因子(CSF)是一类分泌型的糖蛋白,其与造血干细胞表面上的受体蛋白结合,从而激活细胞内信号传导途径,可导致细胞增殖并分化成特定类型的血细胞(通常是白细胞,对于红细胞形成,参见促红细胞生成素)。集落刺激因子的名称来源于它们被发现的过程。在进行造血细胞的体外研究中,发现一些细胞因子可刺激不同的造血干细胞在半固体培养基中形成细胞集落,而这类因子被命名为集落刺激因子。

根据集落刺激因子不同的作用范围,CSF可分为三种,分别命名为巨噬细胞CSF(M-CSF/CSF1)、粒细胞和巨噬细胞CSF(GM-CSF/CSF2)、粒细胞CSF(G-CSF/CSF3)和多能集落刺激因子(multi-CSF,又称IL-3)。它们对不同发育阶段的造血干细胞起促增殖、分化的作用,是血细胞发生必不可少的刺激因子。但从广义上来说,CSF还包括刺激红细胞生成素(Epo)、可刺激胚胎干细胞的白血病抑制因子(LIF),以及刺激血小板的血小板生成素(thrombopoietin)等均有集落刺激活性。

除CSF外,它们的受体在CSF行使功能的过程中也至关重要。不同CSF的受体不同,并且在不同的骨髓群上表达。简而言之,除CSF1R外,每种受体都由特异性CSF结合刺激,而CSF1R可以与CSF1和IL-34结合。

CSF1受体(CSF1R)在单核细胞和巨噬细胞上表达,是唯一一个酪氨酸激酶受体,由CSF1和IL-34激活。CSF3受体(G-CSFR)是中性粒细胞中表达量最高的跨膜蛋白,但它也存在于其他类型的细胞中。CSF2受体(GM-CSFR)在单核细胞、巨噬细胞、嗜酸性粒细胞和嗜中性粒细胞中均有表达。
