OPG是肿瘤坏死因子(TNF)受体超家族的成员之一,又称TNFRS11B或OCIF,属于Ⅱ型跨膜蛋白。OPG主要由骨髓基质细胞表达。此外,在免疫系统(如淋巴结、B细胞和树突状细胞(DCs))和血管系统(如心脏、动脉和静脉)中也检测到其表达。OPG通过与其配体RANKL或TRAIL结合,直接或间接地发挥作用。OPG/RANKL/RANK是OPG的主要信号通路,激活JNK、p38、ERK、Akt和NF-κB等信号分子,主要参与破骨细胞的发生。OPG/RANKL/RANK通路不仅在破骨细胞激活、形成和存活中起主导作用,而且还在其他组织中发挥作用,包括乳腺、大脑和淋巴结。OPG/RANKL/RANK通路的紊乱与人类某些疾病有关,包括绝经后骨质疏松症、类风湿性关节炎(RA)、骨肿瘤和某些骨转移性肿瘤。近年来,OPG作为一种潜在的生物标记物或治疗靶点备受关注。换句话说,OPG抗体或者抗OPG产品在临床研究中的应用有望监测疾病相关的进展和并发症。
