Smo

SMO,即平滑蛋白,别名Smoothened。它是一种细胞表面受体,属于F类G蛋白偶联受体(GPCR)家族成员,是Hedgehog(Hh)信号通路中的关键组分。SMO在细胞内主要通过与Hh配体结合,被激活后能够将信号传导至细胞核,进而激活GLI转录因子,调控相关基因的表达。SMO的来源是人体细胞自身的基因编码,其作用机制涉及到与多种蛋白质和脂质分子的相互作用,尤其是胆固醇的共价修饰对SMO的功能至关重要 。SMO在生物学上的意义非常重大,它不仅参与调控胚胎发育、组织再生和器官修复等生理过程,而且与多种癌症的发生发展密切相关。Hh信号通路在静息状态下,SMO受到Patched(PTCH)蛋白的抑制;当Hh配体结合PTCH时,SMO受到的抑制被解除,从而激活下游信号通路。此外,SMO的异常激活与胶质瘤等多种肿瘤的发生密切相关,因此,SMO成为了抗癌药物开发的重要靶点 。
