MC4R,即黑皮质素4受体”,别名Melanocortin 4 Receptor、Melanocortin Receptor 4、MC4-R、Mutant Melanocortin-4 Receptor等。它是一种膜结合受体,属于黑皮质素受体家族成员,由人体细胞自身的基因编码。MC4R主要在下丘脑中表达,参与控制食物摄取、能量消耗和体重维持等 。MC4R的作用机制主要是通过与黑皮质素激素如α-黑素细胞刺激激素(α-MSH)等内源性配体结合,通过G蛋白偶联的方式激活腺苷酸环化酶(cAMP)信号通路,从而发出饱腹感信号,减少食物摄入。MC4R的激活还涉及多种代谢信号的处理,如瘦素、饥饿素和神经肽Y (NPY)等,调节人体能量代谢和进食行为 。MC4R在生物学上的意义非常重大,它不仅参与了能量平衡和体重调节,还与多种代谢性疾病的发生发展密切相关。例如,MC4R的功能异常或突变与肥胖症有关,MC4R激动剂如setmelanotide已被批准用于治疗某些类型的肥胖症。此外,MC4R的激活还可能影响情绪、奖赏和认知功能,对心理健康和行为也有潜在影响 。
