IL23

IL-23(白细胞介素-23)是一种由IL-23p19和IL-12/IL-23p40两个亚基组成的异源二聚体细胞因子,与IL-12共享p40亚基。IL-23主要通过与其受体IL-23受体相互作用来激活下游信号通路,发挥生物学功能。IL-23受体由IL-12受体β1(IL-12Rβ1)和IL-23受体两个亚基组成,是一种跨膜蛋白。IL-23在机体的炎性免疫反应和IL-17的产生过程中发挥重要作用,主要作用于Th17细胞,促进其增殖与稳定,并诱导Th17细胞产生IL-17A、IL-17F、IL-22等细胞因子,参与多种疾病的发生发展。 IL-23参与的信号通路主要包括JAK/STAT、MAPK和PI3K等信号转导途径。当IL-23与其受体结合后,触发IL-12Rβ1和IL-23R的异源二聚化,激活这些信号通路。IL-23通过RORγt和STAT3等转录因子发挥作用,促进Th17细胞的分化和效应子功能。此外,IL-23还与肿瘤免疫、自身免疫性疾病的发生发展有关,例如通过抑制细胞毒性T细胞在肿瘤组织中的浸润促进肿瘤生长,IL-23R的遗传变异体与多种肿瘤的发生有关,血清IL-23水平的升高与某些癌症的发展阶段相关。因此,IL-23在调节免疫反应、促进炎症反应以及参与肿瘤免疫和自身免疫性疾病的发生发展中发挥着关键作用。

IHC image of CSB-RA011587MA1HU diluted at 1:50 and staining in paraffin-embedded human lymphnode tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.
