IHC image of CSB-RA004935A0HU diluted at 1:100 and staining in paraffin-embedded human lymph node tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

The CD4 recombinant monoclonal antibody is a highly specific antibody against the human CD4 protein. This CD4 antibody was produced in vitro by transfecting human CD4 monoclonal antibody gene- vector clones into the cell line, followed by affinity-chromatography purification from the tissue culture supernatant (TCS). The mice were immunized with a human CD4 synthetic peptide to produce the human CD4 monoclonal antibody. This CD4 antibody has the same isotype as rabbit IgG. It can be applied in ELISA, WB, and IHC.
T cells, natural killer (NK) cells, monocytes, and macrophages all express the CD4 coreceptor. CD4 regulates differentiation, migration, and cytokine expression in macrophages and NK cells. It plays a role in thymic development and antigen recognition in the periphery in T lymphocytes.

功能:Integral membrane glycoprotein that plays an essential role in the immune response and serves multiple functions in responses against both external and internal offenses. In T-cells, functions primarily as a coreceptor for MHC class II molecule:peptide complex. The antigens presented by class II peptides are derived from extracellular proteins while class I peptides are derived from cytosolic proteins. Interacts simultaneously with the T-cell receptor (TCR) and the MHC class II presented by antigen presenting cells (APCs). In turn, recruits the Src kinase LCK to the vicinity of the TCR-CD3 complex. LCK then initiates different intracellular signaling pathways by phosphorylating various substrates ultimately leading to lymphokine production, motility, adhesion and activation of T-helper cells.
