    外泌体是一类细胞分泌的包含了多种RNA、蛋白质以及磷脂等成分的直径约为30-150nm的细胞外囊泡。在细胞培养上清液和体液(乳汁、血清、精液、血浆、唾液、尿液、羊水、脑脊液等)中大量存在。外泌体在细胞间物质和信息传递过程中起重要作用。

    本产品提供了一种简单可靠的方法从细胞培养基上清液、血清、血浆、乳液、尿液、唾液、酵母、植物等样品提取完整的外泌体。本产品提取的外泌体囊泡适用于下游的电镜分析、NTA分析、纳米流式(NanoFCM)分析、Western Blot、荧光定量(qPCR)和高通量测序等应用。

产品评价:尿液所含的细胞外囊泡(EV)可能包含了膀胱癌细胞主动分泌的miRNA,通过检测EV中特定miRNA峰度的变化,可能识别早期癌症。Cusabio外泌体提取试剂盒分离尿液中的EV;一部分EV提取RNA、反转录、qPCR检测内参靶标;另一部分EV简单处理后直接反转录、qPCR检测。CUSABIO试剂盒提取结果不错。

常见问题

1、如何保存纯化获得的外泌体囊泡?
答:短时间内保存于4℃,长时间保存需要放置于-80℃(建议分装后保存、避免反复冻融)
2、如何获得无外泌体血清培养细胞?
答:a、将细胞培养用的血清通过超速离心10h以去除血清中外泌体(无菌操作);
b、可选择无血清培养基进行细胞培养;
c、可选择去外泌体血清进行细胞培养。
3、如何确认所提取到的是外泌体?
答:可以通过特定标志物的WB验证,电镜、粒径(纳米流式、NTA)等方式来确认。
4、与超速离心相比,我们的优点是什么?
答:与超速离心相比,可以简便的、稳定的、高重复性的实现外泌体的回收,且没有仪器设备的限制。
