XPO1

XPO1,即染色体区域维持1蛋白(Chromosome Region Maintenance 1 Protein),也被称为出口蛋白1(Exportin 1),是一种在细胞核输出过程中发挥关键作用的蛋白质。它通过识别并结合含有核输出信号的货物,如某些RNA和蛋白质,将它们从细胞核运输到细胞质中。XPO1的来源是细胞自身的基因编码,它在细胞分裂、基因表达调控、细胞信号传递等方面发挥着重要作用。其作用机制涉及到与Ran GTPase蛋白的相互作用,以及与货物蛋白的结合和释放,从而实现物质的定向运输。XPO1在生物学上的意义非常广泛,它涉及到细胞内多种信号通路的调控,包括但不限于p53信号通路、NF-κB信号通路等,这些信号通路与细胞的生长、分化、凋亡以及对外界刺激的响应密切相关。XPO1的功能异常与多种疾病相关,特别是在癌症中,XPO1的过表达与肿瘤的侵袭性、转移和化疗抵抗性有关。因此,XPO1成为了抗癌药物开发的重要靶点,通过抑制XPO1的活性,可以阻止肿瘤相关蛋白的核输出,进而影响肿瘤细胞的生长和存活。

IHC image of CSB-RA930964A0HU diluted at 1:100 and staining in paraffin-embedded human ovarian tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
