TM4SF1(Transmembrane 4 L six family member 1)是Tetraspanin超家族中亚家族TM4SFs的一员,首次于1986年被鉴定。它具有Tetraspanin家族典型的四次跨膜结构,胞外含两个loop区EC1和EC2,其中EC2区可通过半胱氨酸残基与整合素α5β1结合,影响细胞的黏附和运动。C端胞质区包含PDZ结合域,可与Syntenin-1和Syntenin-2蛋白相互作用介导肿瘤细胞的增殖和转移。 TM4SF1在多种肿瘤细胞表面高表达,包括肺癌、乳腺癌、肝癌、结直肠癌及卵巢癌等,且能与脂质激酶、酪氨酸蛋白激酶等形成复合体,如可结合DDR1,激活PKC和JAK2-STAT3信号通路,促进细胞伪足延伸。初步的研究结果表明,TM4SF1作为具有酪氨酸蛋白激酶活性的单次跨膜受体1(DDR1)的辅助受体,调控下游的PKC/STAT3信号通路,从而影响乳腺癌向肺组织的转移。此外,TM4SF1也是具有新型内化途径的ADC靶点,这意味着它在抗体偶联药物(ADC)的设计和开发中可能具有潜在的应用价值。 于TM4SF1的药物研发处于初级阶段,尚未有成熟的药物应用于临床治疗。因此,对于TM4SF1的研究仍具有深远的意义和广阔的前景。
