TNFR1

TNFR1(Tumor Necrosis Factor Receptor 1),也称为TNF-R55或CD120a,是一种受体蛋白,它与肿瘤坏死因子α(TNF-α)相互作用并在多种生理和病理过程中发挥关键作用。包含一个细胞外域、一个跨膜域和一个细胞内域。细胞外域与TNF-α结合,触发信号传导。细胞内域包含一个死亡结构域(Death Domain),它在启动凋亡(细胞自我毁灭)和其他细胞信号传导通路中发挥关键作用。TNFR1主要通过与其配体TNF-α相互作用来介导多种生物学效应。当TNF-α与TNFR1结合时,它可以引发一系列信号传导事件,包括NF-κB(核因子-κB)的激活和JNK(c-Jun N-末端激酶)通路的激活。在自身免疫疾病和炎症性疾病的治疗中,研究人员已经开发出一些生物制剂,如抗TNF药物(如英夫利麦、阿达木单抗等),用于减轻症状并减缓疾病进展。这些药物通过抑制TNF-α与TNFR1的结合,降低炎症反应。此外,TNFR1还涉及到肿瘤的发展和抵抗性,因此在癌症治疗领域也具有潜在的应用前景。
