    Tumor necrosis factor (TNF)是人体中由TNF基因编码的蛋白质。它是一种细胞因子,能够与TNFRSF1A/TNFR1和TNFRSF1B/TNFBR结合。主要由巨噬细胞分泌,并可以诱导某些肿瘤细胞系的细胞死亡。它是一种强效的致热原,通过直接作用或刺激白细胞介素-1的分泌引起发热,并参与恶病质的诱导。在某些条件下,它也可以刺激细胞增殖和细胞分化。在风湿性关节炎患者中,TNF破坏了调节性T细胞(Treg)的功能,通过FOXP3去磷酸化来实现。它上调了蛋白磷酸酪氨酸酶1(PP1)的表达,PP1去磷酸化FOXP3的“Ser-418”残基,从而使FOXP3失活,导致Treg细胞功能缺陷。

    TNF具有多种生物学功能,包括调节免疫反应、促进炎症反应、参与细胞凋亡等。在疾病发展中,TNF的异常表达或过度激活与多种疾病的发生和进展密切相关。因此,TNF被广泛研究,并成为一些疾病治疗的靶点。

Tumor necrosis factor is a protein in humans that is encoded by TNF gene. Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation. Impairs regulatory T-cells (Treg) function in individuals with rheumatoid arthritis via FOXP3 dephosphorylation. Upregulates the expression of protein phosphatase 1 (PP1), which dephosphorylates the key 'Ser-418' residue of FOXP3, thereby inactivating FOXP3 and rendering Treg cells functionally defective
