SIRT3
SIRT3,作为线粒体中的关键去乙酰化修饰酶,其结构包括一个催化区以及调节酶活性的N端和C端结构。它在细胞应激时转移到线粒体内并成熟,从而发挥其去乙酰化功能。SIRT3主要通过调节线粒体结构和功能,如激活长链脂酰辅酶A脱氢酶,在脂肪代谢中发挥重要作用。在信号通路方面,SIRT3通过调控GSK-3β/β-catenin信号通路来抑制NLRP3炎性小体的表达,从而保护骨骼成骨。同时,在肾脏疾病中,SIRT3通过去乙酰化作用抑制肾纤维化,并调控能量代谢稳态。此外,SIRT3还参与心脏氧化应激、缺血再灌注损伤等生理和病理过程。
