NRP1

神经纤毛蛋白(Neuropilin-1,简称NRP1)是一种单次跨膜、非酪氨酸激酶表面糖蛋白。NRP1最初被发现作为正常胚胎发育、轴突引导和血管形成所必需的神经元和内皮细胞受体,也在参与关键免疫功能的多种免疫细胞类型中表达。现有的研究表明,NRP1在癌症患者的调节性T细胞中升高,提示其可能是癌症免疫治疗的靶点。在COVID-19研究中,人们发现NRP1能够与S蛋白结合增强新冠病毒的感染能力。通过单抗对NRP1进行阻断,实验结果证明,新冠病毒的感染性得到明显下降。另外,NRP1能够与广泛的配体结合,如信号素3A(Sema3A)、生长因子VEGF1、转化生长因子β、血小板衍生生长因子C和D、和C-Met等等。越来越多的文献表明,NRP1是癌症免疫治疗中一种独特的免疫调节剂。在TME中,NRP1内在地调节Treg细胞和CD8+T细胞功能,共同阻碍抗肿瘤免疫。因此,NRP1在肿瘤中作为一个关键的免疫调节剂具有诱人的开发前景。
