LTA基因编码TNF-β蛋白(肿瘤坏死因子β),也称为淋巴毒素α(Lymphotoxin Alpha,LT-α),属于TNSF超家族一员。根据TNF来源的不同,人们将单核/巨噬细胞所产生的TNF称肿瘤坏死因子α;把T细胞产生的TNF称为肿瘤坏死因子(TNF-β)。TNF-β有膜结合和可溶性两种形式,分泌到细胞外的可溶性形式称为LT-α。LT-α主要由活化的CD4+CD8+T细胞、NK细胞、B淋巴母细胞样细胞、骨髓瘤细胞等产生。LT-α在细菌的代谢和致病过程中发挥重要的作用。研究证实在感染结核分支杆菌后,遗传性LT-a缺陷的小鼠比LT-a正常小鼠屯集更多的细菌负荷量。TNF-β除了介导许多炎症、免疫刺激和抗病毒应答。在发育阶段,TNF-β还参与二级淋巴器官的形成,并在凋亡中发挥作用。在TNF-β敲除小鼠中,所有派伊尔氏淋巴集结和淋巴结都无法发育,这表明了TNF-β在免疫系统发育中的重要性。
