LRRC15
LRRC15，即富含亮氨酸重复序列的蛋白15”，别名LIB。它是一种I型跨膜蛋白，属于LRRC超家族成员之一，由人体细胞自身的基因编码。LRRC15在多种肿瘤微环境中的基质成纤维细胞中表达，并且其表达受TGFβ调控。LRRC15的作用机制涉及到与肿瘤微环境中的成纤维细胞相互作用，影响肿瘤细胞的迁移和侵袭，以及可能通过Wnt/β-catenin信号通路促进三阴性乳腺癌细胞的迁移和侵袭。LRRC15在生物学上的意义非常广泛，它不仅作为肿瘤微环境中的间充质标志物，还在肿瘤发展、转移以及对免疫疗法的反应中发挥重要作用。LRRC15的高表达与多种实体瘤类型中抗PD-L1疗法反应不良相关，使其成为癌症治疗的潜在靶点。例如，ABBV-085是一种针对LRRC15的抗体-药物偶联物(ADC)，已在临床前研究中显示出对LRRC15阳性肿瘤的疗效。此外，LRRC15+成纤维细胞的耗竭可以解除对肿瘤浸润性CD8+T细胞的直接抑制，增强其效应功能，并提高对抗PDL1免疫检查点阻断的肿瘤消退。因此，LRRC15是肿瘤免疫微环境研究和癌症治疗中的一个重要靶点。

Immunofluorescence staining of SH-SY5Y cells with CSB-PA819484LA01HU at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
