Kras

KRAS,即Kirsten大鼠肉瘤病毒癌基因同源物”,别名KRAS4A和KRAS4B,这两种变体来源于同一基因的不同剪接形式。KRAS基因编码的蛋白质属于RAS超家族的小GTP酶,是一种在细胞内信号传导途径中起关键作用的分子开关。它通过与鸟嘌呤核苷二磷酸(GDP)或鸟嘌呤核苷三磷酸(GTP)结合,来调节自身的活性状态。在正常细胞中,KRAS受到上游信号如EGFR激活的严格控制,其活性周期性地在激活和失活之间转换,以维持细胞内环境的稳定。KRAS在生物学上的意义非常重要,它参与调控细胞的生长、分化、存活和迁移等关键生理过程。KRAS蛋白通过激活包括RAF-MEK-ERK(MAPK)通路、PI3K-AKT通路以及Ral-GEFs通路在内的多条下游信号通路,影响细胞的生理功能。当KRAS发生突变时,如常见的G12D、G12V、G13D等,它将被锁定在持续激活状态,导致细胞信号传导失衡,促进肿瘤细胞的无序增殖和存活,与多种肿瘤的发生发展密切相关。因此,KRAS是肿瘤治疗中一个极具挑战性和重要性的靶点,针对KRAS突变的靶向治疗正在成为癌症治疗研究的热点。
