FPS1

FPS1,即铁死亡抑制蛋白1,别名Ferroptosis-suppressor-protein 1。它是一种黄素蛋白,由德国发育遗传学研究所和维尔茨堡大学的研究人员发现,FSP1能够作为一种谷胱甘肽非依赖性铁死亡抑制因子。FSP1的作用机制涉及到使用NAD(P)H催化泛醌(CoQ10)的再生,泛醇作为还原形式捕获介导脂质过氧化作用的脂质过氧自由基,从而抑制铁死亡。FSP1的药理靶向作用与GPX4抑制剂可产生协同作用,引发铁死亡。在生物学上,FSP1作为独立的并行系统存在,与GPX4和谷胱甘肽(GSH)协同作用来抑制磷脂过氧化和铁死亡,对于了解铁死亡的病理生理作用以及如何将其用于癌症治疗具有重要意义。

Overlay Peak curve showing HepG2 cells stained with CSB-RA029525A0HU (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
