I purchased item CSB-EP361330IJK lot 03765 and ask the following questions:
1)What is the exact buffer, including pH of each product?
2)What is the sequence of each product including the tags?
3)How was the concentration determined? What is the concentration of each?
4)What is the purity of each item?
CSB-EP361330IJK lot 03765
1) Storage Buffer: 10 mM Tris-HCl, 1 mM EDTA, pH 8.0, 50% glycerol
2) N-terminal 6xHis-SUMO-tag Sequence: (MW: 16 kDa):

Target Protein Sequence (MW: 61.8 kDa):
This sequence has been presented on this page.
3) Concentration: 0.4 mg/ml, by the Bradford Method.
4) Purity: 90%, by SDS-PAGE quantitative densitometry by Coomassie Blue Staining.
I have additional questions about the data that you sent. Would you please help with the questions below?
1). The storage buffer for CSB-EP325878IMI (Flu A) 10% correct? It is not 50%?
2). For Flu A antigen, the buffer contains 0.5M NaCl, while the buffer for Flu B antigen has no salt.

The predicted MW of this sequence indeed is 12.96KD. We forgot to explain to the customer and directly provided 16 kDa MW in the last email.
Actually, the MW of SUMO-tag will be about 4kDa larger. Theoretically, because the isoelectric point of the SUMO-tag is very low, the acid protein will generally run relatively larger.
Combined with our actual situation (statistics of not less than a thousand data), the SUMO-tag is indeed about 4 kDa larger, so please don't worry.
There are also many articles reporting that the SUMO-tag shows relative larger size on gel.
2. About the glycerol content in storage buffer: When proteins shipped, we generally add a final concentration of 50% of glycerol by default. Glycerol has a protective effect on the protein.
First, it can increase the stability of the protein (reducing the polarity of the buffer and preventing protein hydrophobic aggregation).
(This is based on the principle of similar dissolution: polar molecules are easily soluble in polar solvents, non-polar molecules are easily soluble in non-polar solvents,
and polar and hydrophilic (water is a polar molecule) correspond, hydrophobic and non-polar Polarity corresponds to a positive correlation.
Second, the freezing point of the system can be reduced, and the protein can be stored at a low temperature. Especially when the protein is freeze-thawed repeatedly,
protein damage (aggregation or inactivation) can be avoided as much as possible.
As for the final concentration of glycerol, in principle, the final concentration of 5% -50% is OK, may involve the preservation of time.
The preservation time of high glycerol content is relatively longer, but for each delivery, this can adjusted according to the customer's request, there is no problem with glycerol plus or minus.
CSB-EP361330IJK is a in-stock protein, and glycerol content is fixed 50%, already added.
3. Different protein/even if the same protein, the stability in different buffers is not the same, and we will choose the optimal buffer, so the buffers of different proteins are not the same.

功能:Encapsidates the negative strand viral RNA, protecting it from nucleases. The encapsidated genomic RNA is termed the ribonucleoprotein (RNP) and serves as template for transcription and replication. The RNP needs to be localized in the host nucleus to start an infectious cycle, but is too large to diffuse through the nuclear pore complex. NP comprises at least 2 nuclear localization signals that are responsible for the active RNP import into the nucleus through cellular importin alpha/beta pathway. Later in the infection, nclear export of RNPs are mediated through viral proteins NEP interacting with M1 which binds nucleoproteins. It is possible that nucleoprotein binds directly host exportin-1/XPO1 and plays an active role in RNPs nuclear export. M1 interaction with RNP seems to hide nucleoprotein's nuclear localization signals. Soon after a virion infects a new cell, M1 dissociates from the RNP under acidification of the virion driven by M2 protein. Dissociation of M1 from RNP unmasks nucleoprotein's nuclear localization signals, targeting the RNP to the nucleus.
