The recombinant Aquaporin-1/AQP1 was produced as a fusion protein with N-terminal 6xHis-SUMO tagged in vitro E.coli expression system. The encoding sequence contains 2-269aa of human AQP1 protein. This full-length of mature AQP1 protein reached up to 90% in purity, which is determined by SDS-PAGE. It migrated to a molecular mass band of 45 kDa on the gel. The slightly higher result is attributed to glycosylation. And it was also verified with a high activity assessed by its binding ability in a functional ELISA. This recombinant AQP1 protein may be used to synthesize anti-AQP1 antibodies or in the studies of AQP1-mediated signal transduction.
AQP1 is one of the aquaporins, water channels that are critical to regulating water fluid homeostasis in cells involved in inflammation. It exerts a role in macrophage migration and AQP1-mediated some phenotypic and migratory modifications of these cells are crucial for the switch of M0/M2 phenotype. AQP1 localized on lung capillaries may participate in the development of angiogenesis. Particularly, the elimination of AQP1 reduces endothelial cell migration, limiting tumor angiogenesis, and growth. It has been demonstrated that AQP1-expressing tumor cells augment local infiltration and metastatic effects.
