    This rat IL33 ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of rat IL33 in multiple samples, including serum, plasma, or tissue homogenates. The enzyme-substrate chromogenic reaction is also used to amplify the signal and quantify the levels of the analyte through the intensity of the colored product. The color intensity positively correlates with the amount of IL33 bound in the initial step.

    IL33 is released upon tissue stress or damage to act as an alarmin. Different from other IL-1 family members such as IL18 and IL-1β, IL33 is inactivated after caspase cleavage. IL33 binds to its receptor IL-1RL1/ST2 and IL1RAP, activating downstream signaling molecules including MyD88 and TRAF6 ultimately inducing NF-κB, p38, JNK, and ERK pathways. IL33 primarily participates in the induction of type-2 immune responses, eliciting the release of Th2 cytokines thus facilitating allergic reactions. It also supports tissue repair by coordinating the action of innate lymphoid cells (ILCs) and regulatory T cells (Tregs). Additionally, IL33 acts in tissue homeostasis, growth, and immune response. It is associated with the pathogenesis of various diseases, including infection, inflammatory disorders, skin diseases, and various cancers.

    Cytokine that binds to and signals through the IL1RL1/ST2 receptor which in turn activates NF-kappa-B and MAPK signaling pathways in target cells. Involved in the maturation of Th2 cells inducing the secretion of T-helper type 2-associated cytokines. Also involved in activation of mast cells, basophils, eosinophils and natural killer cells. Acts as a chemoattractant for Th2 cells, and may function as an 'alarmin', that amplifies immune responses during tissue injury.; In quiescent endothelia the uncleaved form is constitutively and abundantly expressed, and acts as a chromatin-associated nuclear factor with transcriptional repressor properties, it may sequester nuclear NF-kappaB/RELA, lowering expression of its targets. This form is rapidely lost upon angiogenic or proinflammatory activation.
