Sstr5

SSTR5,即生长抑素受体5,别名生长抑素受体亚型5、SS5-R、Somatostatin receptor type 5等。SSTR5是一种G蛋白偶联受体(GPCRs),主要在垂体中高度表达,参与调控多种激素的分泌,如促肾上腺皮质激素、催乳素和生长激素等。SSTR5的作用机制涉及到与其天然配体生长抑素(SST)或合成激动剂药物如奥曲肽(octreotide)结合,进而抑制腺苷酸环化酶(AC)的活性,通过抑制性G蛋白(Gi)介导,调控胞内cAMP浓度,影响激素的分泌和肿瘤生长等过程 。SSTR5在生物学上的意义主要体现在其作为多种内分泌疾病和肿瘤治疗的潜在靶点。SSTR5的激活可以减少胆囊收缩,增加胆固醇性胆结石的风险,因此,拮抗SSTR5可能会促进胆囊排空,减少胆囊结石的形成 。此外,SSTR5的表达水平与某些癌症的发病有关,例如研究发现SSTR5基因的变异与头颈部癌、肺癌、前列腺癌和肾癌等癌症的发病有关 。因此,SSTR5不仅在调控激素分泌和肿瘤生长中发挥重要作用,也是药物开发和疾病治疗中的潜在靶点。
