Like all recombinant proteins, this Recombinant Human LOXL1 protein was encoded by recombinant DNA. The recombinant DNA was introduced to a plasmid in which the gene of LOXL1 was cloned downstream of a promoter region. When the plasmid was introduced to the cells of E.coli, the E.coli’s own protein synthesis pathways would then result in the expression of the LOXL1 protein. And the next step was protein purification. The purity of this recombinant protein is 90%+ determined by SDS-PAGE.
LOXL1 belongs to LOX family proteins. Members of the LOX family are secreted by tumors and are the subject of extensive efforts aimed at understanding their roles in cancer. For instance, LOXL1 can protect glioma cells from anoikis and IR stresses, rendering cells resistant to apoptosis. In detail, LOXL1 is upregulated by the VEGFR-Src-CEBPA axis in suspended glioma cells, and LOXL1 and BAG2 proteins can interact in glioma cells, preventing BAG2-K186 ubiquitylation depending on LOXL1 enzymatic activity and stabilizing BAG2 to ultimately promote cell survival. Besides, the aggregation of the LOXL1 protein coupled with age-related decline in cellular proteostasis capabilities may result in a build-up of LOXL1 containing protein aggregates. -

Sorry, as we haven't tried this buffer before, we don't have raw material for these two kinds of buffer, is it possible for you to accept any other buffer that without any EDTA and DTT ?The final buffer will be determined during the manufacturing process.
We produced CSB-EP013040HU recently, the buffer is 100 mM Tris-HCl, 0.4 M Arg, pH 8.0, 20% glycerol,we can send the COA for your reference.

We really want the active form of this protein.I know that the lab does not test or QC for any kind of activity, but I'm still interested. Plese check the following note:
LOXL1 is an amine oxidase and its activity depends on a coordinated copper ion, therefore, having Tris (contains primary amines), EDTA and DTT might inhibit or kill its activity. Presumably, any buffer system other than Tris w/o EDTA and DTT should work however previous literature suggests that these enzymes have the best activity in Sodium Borate buffer, pH8.2.
Do you guys have any suggestions about another buffer?
Yes, we can't guarantee the activity. But we can try to prepare the protein according to your requirement.
We will purchase sodium borate to have a try, and provide the protein in the buffer of Sodium Borate buffer, pH8.2,
If the protein is not stable in this buffer, we will communicate with you in advance, if you finally accept the other buffer such as PBS, HEPES, CB buffer after the Sodium Borate buffer trial is failed,there isn't any extra charges for Sodium Borate buffer trial.
But if you don't accept any other buffer after the Sodium Borate buffer trail fails, there will be some extra charges.
The extra charges depends on which expression system you choose and which size you need.
